Where can I buy sheets of acrylic in Manhattan?
I want sheets of clear acrylic .25″ thick, around 12″x12″, maybe larger. I’ve tried the art supply shops (Dick Blick, Lee’s, Pearl Paint) but none of them have it in store.

There are several hardware stores (Lowe’s and Home Depot) in your area. The item you asked for is also known as Plexiglas. Most chain hardware stores will cut it for free. It comes in 8′ x 12′ sheets but you can buy any amount you need, just ask a customer service rep to cut it.
